Abstract:
Non-enzymatic protein liqueries are a type of post-translational modification that occurs as a result of a direct chemical reaction between active acyl donors, such as acyl-CoA, and nucleophilic groups in proteins without enzymatic intervention. This phenomenon has received increasing attention due to its potential role in linking the metabolic state of the cell to the functions of proteins. Despite its importance, the mechanical understanding of non-enzymatic fluid interactions is still limited, especially with regard to how environmental factors within the cell affect their behavior Interactions. Most previous studies have focused on descriptive aspects or enzymatic mechanisms, while the kinetics and specificity of these reactions under physiological conditions are still not sufficiently clear. Therefore, this research aims to develop an integrative conceptual framework that links intracellular conditions, such as pH and acyl-CoA concentration, and between the dynamics and selectivity of the interaction. The research analyzes how changes in the cellular environment affect nucleophilia, reaction rates, and the likelihood of modification at specific sites on proteins. The results suggest that non-enzymatic questions are not entirely random, but are influenced by the interaction of chemical agents and local protein environments that lead to preferential patterns of modulation under certain metabolic conditions. The results also showed that increased acyl-CoA availability and pH change are key factors in enhancing reactivity and influencing the distribution of modified sites. Overall, this research demonstrates that non-enzymatic questions are a process dependent on cellular context and reflect the interaction between metabolic status and protein chemistry, providing a more comprehensive understanding of their functional role within biosystems.
